Mutants of Yeast Deficient in Cytochrome C.

چکیده
YTOCHROME C is an ideal protein for biochemical studies because of its ease of isolation and low molecular weight. Its primary structure from a number of species is entirely (cf. SMITH, MATSUBARA, MCDOWALL and ROTHFUS 1963) or partially (cf. TUPPY 1958) known. Therefore, any system exhibiting genetic control of cytochrome c would be of considerable interest for investigating geneprotein relationships. In a previous investigation, a mutant of yeast was shown to have a decreased concentration of cytochrome c controlled by a single gene. The cytochrome c isolated from this mutant was found to differ from wild-type cytochrome c by a number of criteria indicating differences in primary structure ( SLONIMSKI and SHERMAN, in preparation). The investigation reported herein deals primarily with the isolation and genetic analyses of other cytochrome c deficient mutants.

The mutational alteration of the primary structure of yeast iso-1-cytochrome c.
A benzidine-HzOz staining procedure and genetic tests were used for the detection of cytochrome c-deficient mutants of the cyl, cy2, cy3, cy4, and cy6 loci. The chromosomal gene, CY1, was shown to encode the primary structure of iso-l-cytochrome c from bakers’ yeast. Sixteen cyl mutants were isolated after examining over 1 million colonies. These cyl mutants could be categorized as follows: (a)...
متن کاملStructural Characteristics of Stable Folding Intermediates of Yeast Iso-1-Cytochrome-c
Cytochrome-c (cyt-c) is an electron transport protein, and it is present throughout the evolution. More than 280 sequences have been reported in the protein sequence database (www.uniprot.org). Though sequentially diverse, cyt-c has essentially retained its tertiary structure or fold. Thus a vast data set of varied sequences with retention of similar structure and fun...
متن کاملProduction of Ice Nucleation Deficient (Ice-) Mutants of the Epiphytic Strains of Erwinia herbicola
To mutate the Ice Nucleation Active (INA) gene in Erwinia herbicola strains, Tn-5 transposon carried by Psup2021 plasmid was used. This plasmid was transferred to the bacterial cells by electroporation. Electrotransformation was carried out for 2.5 ms at 1800 v and 1 mm distance between the electrodes. Polymerase chain reaction was used for determination of presence or loss of INA gene, using a...
متن کاملCardiolipin Content of Wild Type and Mutant Yeasts in Relation to Mitochondrial Function and Development
The phospholipid composition of various strains of the yeast, Saccharomyces cerevisiae, and several of their derived mitochondrial mutants grown under conditions designed to induce variations in the complement of mitochondrial membranes has been examined. Wild type and petite (cytoplasmic respiratory deficient) yeasts were fractionated into various subcellular fractions, which were monitored by...
متن کاملGenes affecting the expression of cytochrome c in yeast: genetic mapping and genetic interactions.
The four mutant genes, cyc2, cyc3, cyc8 and cyc9, that affect the levels of the two iso-cytochromes c in the yeast Saccharomyces cerevisiae have been characterized and mapped. Both cyc2 and cyc3 lower the amount of iso-1-cytochrome c and iso-2-cytochrome c; whereas, cyc8 and cyc9 increase the amount of iso-2-cytochrome c. The cyc2, cyc3, cyc8 and cyc9 genes are located, respectively, on chromos...
